Abstract
The cell cycle is a fundamental process in development and the maintenance of homeostasis. Various proteins regulate the cell cycle, such as Y-box binding protein -1 (YB-1). YB-1 transcriptionally regulates cell cycle proteins such as cyclin D, E and A. In addition to this, knockdown of YB-1 has been shown to be associated with cytokinesis failure. Data from our lab suggest that YB-1 also interacts with various cytoskeletal proteins, such as actin and tubulin, to regulate the cell cycle post translationally. The aim of this study was to visualize the relative location of YB-1 and FLNB during the cell cycle and to document the effect of YB-1 knockdown in cell cycle progression.
We have visualized the cellular location of YB-1 and FLNB during mitosis in fixed A549 cells. We have also visualized the cellular location of YB-1 and FLNB in live cells using live cell imaging. We have also determined the effects of YB-1 knockdown with regards to cell cycle progression. We have determined that YB-1 knockdown results in cell cycle inhibition and accumulation of cells in G1. Finally, we have shown that FLNB and YB-1 share similar cellular location, laying the foundation for further investigation.
